Scientific Abstract

By combining top-down fabrication methods and bottom-up strategies, we will generate a 3D ex vivo model of the glomerulus. To do this, we will first investigate the behavior of glomerular endothelial cells and podocytes in a co-culture model on an artificial basement membrane. We will then structurally mimic the geometry of the glomerulus by specifically integrating mesangial spheroids between capillary loops consisting of endothelial cells, artificial basement membrane and podocytes (bioassembly). In this way, we want to investigate the effect of the 3D structure on self-organization, maturation, paracrine cell interaction and formation of extracellular matrix.
