Rheological Analysis of Saliva Samples in the Context of Phonation in Ectodermal Dysplasia

Abstract

Introduction: Ectodermal Dysplasia (ED) is a group of rare inherited disorders affecting embryonic ectodermal structures, characterized by reduced development of skin appendages and certain eccrine glands. As many affected males display reduced salivation and impaired acoustic quality, ED offers a unique chance to study the fundamental role of laryngeal mucus in the phonation process. The aim of this study was a thorough rheological analysis of saliva samples serving as non-invasively obtainable substitute for the laryngeal mucus.


Methods: Saliva samples from 36 ED patients and 99 control subjects were collected for 5 min following a 15 min abstention from eating, drinking, or smoking. Rheological measurements employed a plate-plate rheometer (plate diameter: 25 mm; slit width: 120 μm; 25°C). Frequency sweeps spanned 1 Hz to 10 Hz (15 steps; shear stress τ = 0.1 Pa) and shear stress amplitude sweeps varied from 0.01 Pa to 20 Pa (30 steps; f = 0.5 Hz). Storage (G′) and loss (G″) moduli were recorded and compared between ED and controls.


Findings: ED subjects exhibited significantly lower G′ and G″ at lower frequencies and strains, yet slightly higher values at increased frequencies and strains among males.


Conclusion: These findings suggest reduced resistance of saliva to external forces for ED. Transferred to the laryngeal level, this might impair the mucus’ retention rate on the vocal folds. The results also indicate an altered hyaluronic acid content and the underlying molecular mass distribution in ED saliva, guiding further investigations of voice conspicuities.
